Non familiar aggression

I'm looking for advice. My son has 2 cats, sister and brother. They are around 4 yrs old and hes had them from kittens. The female has non familiar aggression and hisses as anything or anyone unfamiliar. She doesn't attack though but makes sure you stay away. Her brother has had to go for overnight stay at the vets and came home yesterday. The female is beside herself, hissing, tail like a bottle brush and will not let my son or her brother near her. My son had to lock her in another room as the male is convalescing. He's tried feliway, stroking the male then stroking the female. Nothing helps. He is really worried. Has anyone got any advice?
